Hi everyone I'm Jack Red, here's a fight for the longest time I personally didn't want to see. More recently around a few months ago, all of that changed. Now I really would love to see Death Battle do this fight. Two legendary FPS heroes that can handle armies mostly on their own. Sure it's another original vs. rip off fight but it will be one of the craziest in that regard.
Duke Nukem
No no not that Duke Nukem, this Duke Nukem!!!
Duke Nukem took on Dr. Proton's robot army and two alien armies to protect Earth. This guy did enough damage to force the aliens to come back a little over a decade to attempt finishing the job. By this point, Duke has become a huge celebrity with many things under his ownership. Duke is a man you really do not want to piss off.
Serious Sam
Sam Stone better known as Serious Sam takes on the Mental's army. Virtually endless amount of alien creatures that are more attack than think of strategies. At one point, Serious Sam was the only human left standing against such a threat. Trust me when I said you never heard of an army this mess up in your entire life.

Duke Nukem
Duke is known to be arrogant to a fault. He doesn't take orders well from anyone else except the general. He really doesn't like having his babes getting kidnapped by aliens. Let's be honest Duke is a big pervert. He goes in usually without much of a plan. Some of his own weapons can do fatal damage to himself if he isn't careful.
Serious Sam
Serious Sam can be arrogant to a degree. He prefers blowing stuff up than to figure out a complex puzzle. Even he states how much he loves them lol. You will be very surprised he doesn't get much time to himself since going back in the past. Technically speaking he should be already pushing his limits from the constantly waves of enemies.
Duke Nukem
Advantages
+Both Shrinker & Expander will be very difficult to avoid
+If given Duke 3D Freezethrower, each projectile will bounce off it's surrounding
+Took down three armies
+More confirmed history out of the two
+Technically has more going for him non canon side of things
+At least he got a little over a decade break
+Not as bright as Sam
+Doesn't rely on orders as much as Sam does
+More vehicles that could be used in the fight

-Ever since going back to the past, Sam haven't got a solid break from fighting Mental's army

Verdict
Before Duke fans gets upset please try listening to why I'm picking Serious Sam first. Obviously I did leave out weapons for both characters could use in a fight. Honestly it wouldn't even matter because even giving Duke his best arsenal set. Let's face it, the man is really out gunned here. I mean come on, Serious Sam has more variety of weapon classes alone.
For example, Duke uses mighty foot while Sam could use his knife or chainsaw. Even in a shotgun to explosive battle, Sam still has the advantages. Hell some of the weapons has a very high ammo amount greatly trumping Duke's entire arsenal. Non canon side of things, Sure Duke does have a slightly better shot at winning.
Until you play Serious Sam Double D or it's updated version Double D XXL. Where Sam can wield six weapons at once. Yes you can have multiple of the same weapons. The fact Sam has that non canon would make this fight even more not fair. Sam also can get golden weapons to greatly reduces Duke's chances of winning.
Trust me even defeating a brain washed Santa Claus won't help him here. Duke does get breaks in between the main games. In fact both Duke Nukem Forever & the fifth episode of Duke 3D. He had several years to do what he wants until the aliens returned. Sadly Serious Sam never got a solid break in the distant past.
At first this sounds like Duke could take advantage of. However this really isn't the case at all. This guy held his own against massive hordes of enemies with possibly thousands in the body counts alone. Which easily trumps the combine armies Duke himself took down. Also two of them at one point was fighting each other while trying to hunt Duke down.
Obviously Duke is facing smarter overall enemies while most of Serious Sam's are mindless creatures. Despite this, Sam did take down some of Mental's greatest allies. All of them commanded a lot of his army with some form of intelligence. In fact they all had centuries of a few thousand years under their belt. Sam has a stronger track record of defeating giant creatures.
The biggest Duke has taken down in canon was Alien Empress. It even states in the dlc campaign, if she gives birth. The alien will have enough in numbers to wipe out the rest of humanity. Essentially this was the alien of the highest order. Sure Duke had to rely on a moon vehicle for a good amount of it.
Serious Sam has defeated most of them on a regular basis to begin with. Most giant bosses he took down without having much at all. A great example is both of them had a giant alien spacecraft boss fight. Duke relied heavily on turrets while Sam had just enough ammo to take one down. Keep in mind he stayed on the ground while Duke was high up on an building.
Power ups wise this is no contest folks. Even the best of Duke's simply can not compare with what Sam does. He could become temporary invisible or invulnerable. Duke wouldn't have much counters for either. By default, Sam moves faster than Duke using steroids. So when he gets Serious Speed, he becomes too fast for Duke to come up with.
I should mention Sam is ultimately taking on a god. Duke never faced anything similar to that. In fact most of Sam's enemies would be a new ball game for Duke while most of Duke's enemies are similar to his own. For crying out loud, Sam has to stay on top of kamikazes that usually come in big numbers trying to blow him up.
He can faces ramming animals that doesn't slow down much even running into a wall. I didn't even talk about their respective game changer weapons yet. For Duke, his greatest chances of winning is using either Shrinker or the Expander. He only needs one shot with Shrinker to shrink Sam down to being stomp on.
Even if Sam were to take a hit I'm positive he can fight Duke off long enough to return to normal size. The real question is constantly how fast the Expander fires. Could Sam move fast enough to avoid too much exposure to causing him to grow too big to explode? I believe the answer is a solid yes he can.
On Sam's side of things there's the Cannon. Sam's answer to weapons like Doomguy's BFG I'm really not kidding. Sure it doesn't look like much but this thing can guarantee wiping out an massive horde of enemies in record time within a few shots. Yes the cannon balls sometimes explodes doing damage to enemies.
The last major reason Sam would win this fight should've been obvious by now. His driven allows him to adapt to any situation much faster than Duke does. As stated before he kept facing hordes of enemies with little to no break ever since arriving in the past. Duke only time traveled in canon to the future to stop Dr. Proton.
Obviously facing in the past greatly affects time way more than fighting in the future. A perfect example of Sam winning this has to be taking on Mental's home. He took on a ridiculous amount of enemies before facing Mental inside his home turned into a gigantic tank. This alone throws even the best Duke feats away alone.
In short, Sam has more of most weapon classes. He has defeated way more enemies than Duke does and simply more adaptable in an intense combat situation overall. I do respect both characters with an passion now. I got to be honest this will still be fun to watch but quickly becomes a stomp of a fight. Regardless it better be done in 3D.
